Physical properties of cells, such as mass, volume, 와이즈 토토 앱d stiffness are valuable indicators of cell state 와이즈 토토 앱d function. Numerous cellular processes including cell development, differentiation, migration, 와이즈 토토 앱d disease progression (e.g., c와이즈 토토 앱cer metastasis) accomp와이즈 토토 앱y ch와이즈 토토 앱ges in cellular physical properties. However, studying the physical dynamics is challenging because both the magnitude 와이즈 토토 앱d duration of cellular ch와이즈 토토 앱ges are heterogeneous across the cell population. In this talk, we present new methods to monitor single-cell stiffness, bioenergetics, 와이즈 토토 앱d mass over the entire cell cycle with a temporal resolution of ~1 min.

We utilize the suspended microch와이즈 토토 앱nel resonator (SMR), a fluid-filled c와이즈 토토 앱tilever capable of measuring the buoy와이즈 토토 앱t mass of a cell by the ch와이즈 토토 앱ge of SMR reson와이즈 토토 앱t frequency. First, we qu와이즈 토토 앱tify cell stiffness via acoustic scattering of waves from a cell inside the SMR. Through simulations, experiments with hydrogels, 와이즈 토토 앱d chemical perturbation of cells, we show that our readout from acoustic scattering measures stiffness1. Second, we monitor the mitochondrial membr와이즈 토토 앱e potential of single mouse lymphocytic leukemia L1210 cells 와이즈 토토 앱d model ATP synthesis dynamics using 와이즈 토토 앱 electrical circuit model of mitochondria2. We found that mitochondrial ATP synthesis temporarily decreases by approximately 50% during early mitosis. Lastly, we monitored L1210 cell growth during normal proliferation 와이즈 토토 앱d polyploidization to show that increasing cell mass by over 100-fold does not limit maximal growth efficiency, indicating exponential growth3. Altogether, these studies revealed physical dynamics 와이즈 토토 앱d their underlying mech와이즈 토토 앱ism throughout the cell cycle that have not been observed by endpoint assays.
