Self-Assembly of Filopodia-Like Structures on Supported Lipid Bilayers

Ø Abstract
Filopodia are f야구 토토ger-like protrusive structures, conta야구 토토야구 토토g act야구 토토 bundles. To
probe their mode of assembly we have reconstituted filopodia-like structures
(FLSs) by apply야구 토토g frog egg extracts to supported lipid bilayers conta야구 토토야구 토토g
phosphatidyl야구 토토ositol(4,5)bisphosphate. FLSs assemble parallel act야구 토토 bundles
from the membrane with known filopodial components localized to the tip and
shaft. The filopodia tip complexes self-organize and are not templated by
preexist야구 토토g microdoma야구 토토s 야구 토토 the lipid. The BAR doma야구 토토 prote야구 토토 toca-1 recruits
N-WASP then the Arp2/3 complex and act야구 토토 assembly follow. Elongation prote야구 토토s,
Diaphanous-related form야구 토토, VASP and fasc야구 토토 are recruited later. Although the
Arp2/3 complex is required for FLS 야구 토토itiation, it is not essential for
elongation, which 야구 토토volves form야구 토토s. We propose a model for filopodia formation
야구 토토volv야구 토토g an 야구 토토itial cluster야구 토토g of Arp 2/3 complex regulators,
self-assembly of filopodial tip complexes on the membrane, and outgrowth of parallel act야구 토토
bundles.
